The Li lab at Memorial Sloan Kettering Cancer Center studies how the immune system perceives cancer and the mechanism of cancer immunosurveillance.

In this talk, Li discusses the MMTV-PyMT mouse model of breast cancer, in which tumor growth triggers a granzyme B response. Tumor-associated granzyme B-expressing innate lymphocytes are type 1-like ILCs. ILC1ls and ILTC1s are tissue-resident cells. IL-15 is essential for the generation of ILC1ls and ILTC1s. IL-15 is constitutively expressed in mammary epithelial cells and mammary tumor cells. Ablation of tumor cell-produced Il-15 results in accelerated tumor growth. Enclosed by extracellular matrix and basement membrane, ILC1ls and ILTC1s are in contact with luminal epithelial cells. Transformation of mammary luminal epithelium results in local expansion of ILC1ls and ILTC1s. ILC1ls and ILTC1s mediate cancer immunosurveillance in vivo.
