There are 40 prayer beads in Sekiro. You need 4 prayer beads to make a prayer necklace which will increase your posture and vitality meter...

Two of the prayer beads are easily missable and can only be obtained when going for the "Purification Ending"...

To get the Purification Ending, you will need to side with Kuro and defeat the Owl.
Then go to Isshin's room (follow the smoke signals on Ashina Castle roof) and eavesdrop on him and emma.
Rest and then eavesdrop on Kuro in his room.
Then talk to Emma about Kuro.
Rest and talk to Emma again then warp to the "Old Grave" idol and go left of it and you will see Emma talk to her.
Then warp to the "Dilapidated Temple" Talk to both Emma and the Sculptor and then eavesdrop on them.
Talk to Emma again and ask her about what you heard and she will give a bell use it at the Buddha Statue and you will at the Hirata Estate once again...
