I wouldn't call stuff Best in Slot (your goal is probably different from mine), because I wouldn't want to setup proxy detonation on every build or use Thrassian Stranglers and gamble for crits out of stealth, I'll just call these schematics full damage practical setups.

Instakill - burst that occurs at same time or at least a combo that all happens in less than or equal to a second
Otherwise It's a combo that involves CC be it from attacking from stealth or using a cc ability. Usually more damage drawn out but reliant on target not being on CC immunity.
