Find and kill the three targets within three minutes without being detected. The three targets are somewhere within the green circles on the map. Their locations seem to be fixed, and they're all quite close to each other, so this is an easy one. Start by heading east along the rooftops, past the group of Thieves, and look down from the south end of the rooftops with Eagle Vision active to spot one of the targets marked in gold walking below. Once you see him, climb down, walk up behind him and use your Hidden Blade to end him.
Make your way to the small bridge east of the first target's location. The second target should be walking on this bridge. Ice him with the Hidden Blade and then continue east to the next green circle.
