Who?
When starting a new game of The Binding of Isaac Rebirth - The only playable character you are presented with in the beginning, is Isaac himself. Though there are many more to be unlocked and explored.

What?
There are eleven playable characters in The Binding of Isaac: Rebirth in total. Five of these characters (Isaac, Magdalene, Cain, Judas, and ???/Blue Baby) came with the original Binding of Isaac and its following updates and expansions. While Rebirth contains four new characters (Azazel, Lazarus, Eden, and The Lost.)
Why?

Each Character have different base stats which consist of Damage - Tears - Shot Speed- Range - Speed - Luck and Different Starting Pickups and Items.
All of these things together, can give each a unique play style such as Lazarus reviving when killed or Azazel having the ability of flight and no tears at all. In fact Azazel starts with the ability Brimstone and short range to cut its effect, however, if you were to gain the item brimstone, it would give him the full ability. So outside of primary stats, each charcters stats can be replicated with the same items, each of which is located on the wiki. A great example is
With a dice room which will randomize your items, even Azazel can be grounded and will gain tears.
Or even Eden who gains random items and a random amount of hearts to start with.

Appearance wise they all appear to use Isaac as a base template, as if his Mothers religious Pyshosis over time has had a tangible effect on him and he reverts into these characters as a form of escape. Magdalene is clearly Isaac in a wig.

Not surprisingly Most of the characters names, are biblical references - Eve being derivitive of Adam and Eve or Cain and being a reference to Cain and Abel.




Charcters also have their own sets of achievements which only they can specifically unlock.
Where/When
Some unlocks are fairly easy, such as Eve who is unlocked by not picking up any heart containers for two levels, you can even just do that on a run and quit out to still get the unlock.
However some won’t give you any clues at all, such a Eden , who just shows as a treasure chests which says locked. To unlock Eden you need to
Every time Mom's Heart or It Lives is beaten afterwards with any character one Eden Token will be obtained. Each time a game is started as Eden one of such tokens is consumed. All of this info is present on the wiki with in depth detail as to how to unlock characters including hidden ones which don’t show at all like The Lost which is near impossible to “stumble ‘ across his unlocks s you have to die consecutively with certain characters no other deaths or closing the game in between runs… so yeah, never going to figure that out by myself.
