53This video guide tutorial shows the locations of all 36 heart squids in Shantae & the Seven Sirens and what powerups you need to get them. Heart squids are pieces of a heart container. Once you get 4 heart squids, they can be melted down at the Squidsmith’s for a heart holder to increase Shantae’s max health by one heart.
This guide isn’t organized by the order in which you can get the heart squids. It’s organized by location for easy reference. I tried to mark the points on the map where the heart squids were located and show what fusions/dances were needed to obtain them. There are multiple ways to get these. This is just what I did.

Island East / White Map Area – Right of Arena Town

Jump on boxes - watch spikes. You can use the Newt to stick to the sides of boxes & wait until spikes disappear to jump on top of them.

Dash Newt over to the platform, then Jet Octo to jump. Repeat until you get to the top. Dash over. Watch the spikes.

Refresh Dance to purify poisoned water. Sea Frog to swim
Sea Vent Lab / Laboratory Dungeon

Go into box, get sent flying as Bonker Tortoise to break through wall

Bonker Tortoise to break through wall & go into box pointing up to break rocks above. Try not to fall back down when you un-fusion

Carefully fall from the top right corner of the map, near the boss dungeon. Watch spikes. You can use the Dash Newt to stick to the wall (if there are no spikes) to stop your descent & drop down. You could also just lean to the left as you fall to land on the platform. Go into the room & to the box. Go left, then down, then left. This will send you as the Bonker Tortoise through most of the level until you reach the squid.
The Boiler / Sunken Shipyard Dungeon

Ride on the eyeball boxes (they go in whatever direction the eyeball is looking). Go right, then up, then left, then left again and jump

Ride the eyeball box up – stay on until you reach the very top, and then jump
Laboratory / Light Blue Map Area

Swim up as the Sea Frog

Spark Dance to get ropes to fall. Jump across to the squid. I used the Dash Newt, but it’s not necessary.
Tour Route / Purple Map Area

It looks like there’s nothing there, but a platform will appear if you walk across. You can use the Seer to see the platform, but it isn’t necessary.
6:15 Squid 31 Mining Site
Use a combo of Jet Octo & Dash Newt to get across the gaps w/o hitting the spikes. Bonker Tortoise to break the rocks. Be careful to un-fusion so you don’t fall. Dash Newt to grab the wall, then dash to the squid.
