The strategy for perfect M1 score is to defeat all 5 tentacles and Qliphot Roots in SSS letter.

For standard tentacle HP (130), you get 27000 after taking out the 5 tentacles. And after the boss fight, Stp value is 11932.
However, in this run I get 26669 Stp for the tentacles, and 12076 Stp after the boss.
The fact that the intermediate score is lower, but the final one is higher may be puzzling, but here's the explanation:

- Stp calculation: ((Defeat pts) * 100 + (E-SSS pts)) / (Total damage)
- E-SSS pts: Damage * Stp factor * Letter factor

"Defeat pts", "Stp factor" and "Letter factor" are constant values. "Damage" and "Total damage" are equal. Since we know Stp = 26669, we can resolve for damage. Let X be damage for all 5 tentacles:

((Defeat pts) * 100 + (X * Stp factor * Letter factor)) / X = ((5 * 3900) * 100 + (X * 3 * 8000)) / X = 26669;
1950000 + 24000X = 26669X;
X = 1950000 / 2669 = 730.6;

So, Damage for all 5 tentacles is around 730.6. This value is just an approximation, as the 26669 Stp were rounded down -you can plug Damage back into the formula for verification.
Damage for 5 standard HP tentacles should be 5 * 130 = 650, so there's an extra 80.6 damage points one or more tentacles are taking.

We can see how this increased damage affects the final Stp calculation, which also takes the Qliphoth Roots fight into account:

((5 * 3900 + 90000) * 100 + (730.6 * 3 + 6000 * 1.1) * 8000) / (730.6 + 6000) = 12076 Stp

In summary, even though Stp for the tentacles is lower than the expected 27000 Stp, the final Stp goes above 11932 because the increased damage for the tentacles gives their fight more "weight" in the final calculation.
It's also worth noting that for only one type of enemy, the lower the damage points the higher the Stp (Vergil glitch).

Not really sure how to extract more damage points for tentacles, I just oserved that Payline attacks seem to add more than 130 damage. It could also be hitting the spike for all I know.

Maybe it'd be possible to farm even more Stp by letting the Green Empusas heal the tentacles.

Remarks:
- After the checkpoint, I wait for Red Empusa to escape, as this allows the second Green Empusa to spawn. It's more convenient to grab both Green Empusas for EX-taunt, as once you let them go, they're both stunned for a few seconds. Otherwise, the Green Empusa may get in the way trying to kill the tentacles and take damage, thus lowering the score.
- After taking out the tentacles, I get away from the fight so I can see my Stp.
- During the boss fight, if the Antenora human shield takes damage, the final score also goes down.
