In this lecture we discussed the concepts behind Michaelis Menten kinetics with consideration of an example of penicillin as substrate and beta lactamase as enzyme. We discussed the different parameters of Michaelis Menten kinetics like Michaelis constant (km), maximum velocity of reaction (Vmax) and turnover number of enzyme ( Kcat) in details.
We discussed the concepts of Km , Vmax and Kcat properly. We came to Know that what Vmax actually means and how Vmax is dependent on enzyme concentration along with units of Vmax. We also understood the concept of Michaelis constant and what it actually means. How Km relates affinity of enzyme with substrate. What is Kcat or turnover number and how is it different from Michaelis constant Km.
We also discussed the original concepts of Michaelis Menten kinetics and how Michaelis Menten kinetics is important in terms of enzyme kinetics.
What are the different parameters in Michaelis Menten equation and how graph is plotted for given data.
