It is possible to do a point of damage and get all five of the blue splotches in one go, but you need to be basically perfect and it is not necessary to get the XS. I did manage to do it once in this video, though.

If you're facing a partially damaged core, you're going to want to laser the core first. That way, the soma is respawning while you're draining the blue splotches, so you waste less time. This rule doesn't hold for the last time you damage it, since it doesn't have to respawn, and if you drain the blue splotches first, you end up with less vital deamage, so you'll need to fix less at the end.
