You can't access this room until you get double-jump. The problem is that the the purple jewel you place to open the trap door can get stuck, so that it stays closed instead of opening. Even after you work your way back up from the three treasure chests, the trap door might not open. The solution is to bring up the options and quit back to the main menu. You will lose your progress since you opened the "special" chest, but the trap door should now be in the open position, so you can get out and the only thing you need to do is loot the other two chests before leaving.
