Jinzaemon fight begins at 1:52 & Doujun at 4:42, fought separately in NG+1 without Kuro's Charm.
This is the more direct method, but the quickest way to deal with them is to use Gachiin's Sugar / Spiritfall and sneak up behind them for an easy deathblow. After giving Doujun the red eyes, exhaust his dialogue and reset the area. You'll notice Jinzaemon will be missing from his cell and you can then go to the island to fight him. If you continued with Doujun's dialogue (i.e. eavesdropped, reset and spoke with him), he'll end up on the island with Jinzaemon and you'll have to fight them both.

For Jinzaemon, stay close and attack. Like Samurai Generals, if you move too far away he'll go into a hunched pose, quickly walk towards you at the last second and perform a strong attack that deals a ton of damage. He has a couple of slower attacks at close range that require more accurate deflection timing (or tap the guard button twice and you might get lucky). Also, it may seem spammy but if he deflects your attack, you need to deflect immediately regardless of whether you see him attack you. It's safer to do this as he can perform a left-to-right slash that comes out quickly - possibly quicker than you can deflect on reaction.

Doujun is much easier, you just need to get in close and attack until he deflects, and then watch his hands. He almost always counters with a two-handed punch - time your counter-deflection when you see his hands move and then immediately attack again to interrupt his follow-up.

There's an odd bug with Jinzaemon Kumano and a possible stun-locking opportunity against Doujun (not 100% sure). Description below, video demonstrating both is here: https://youtu.be/LAbsTsrg8Mk

When you fast-travel to the Abandoned Dungeon, you'll hear the encounter music and I think that's because the Ashina soldiers are briefly alive when you get there. Jinzaemon and Doujun are set to be aggressive against those soldiers, and rarely you might see Jinzaemon attack the dead soldiers on the island as you approach him. You can just walk up behind him and deathblow him when this happens.

For Doujun, I don't think he ever blocks your first attack. If you slow your attacks down to "reset" his guard instead of mashing the attack button, it might be possible to stun-lock him and slowly R1 tap him to death. It could just be coincidence, but he's easy enough and I haven't tested this any further.
