Sergeant Joseph Turok (Gregory Cruz), along with other members of the Whiskey Company, are tasked with capturing General Roland Kane (Powers Booth), a war criminal who resurfaces on a remote planet after years of disappearance. Unbeknownst to the Whiskey Company, the planet is controlled by Mendel-Grumman (MG), where Kane is the commander of the MG military. Kane is also the former leader of the Wolfpack, a special operations unit. Turok is a member of this unit but abandoned its ideals during a mission in Colombia. Slade (Ron Perlman), a member of the Whiskey Company, holds a grudge against Turok, as his brother, also a Wolfpack member, was killed during the mission.

As they approach, the Whiskey Company is shot down and makes an emergency landing on a genetically modified planet, scattering the group. The land is infested with genetically modified dinosaurs that the MG created to be weapons. Turok finds Sergeant Major Henderson (Steve Van Wormer), who was killed by the Utahraptor, then Slade. The two encounter the gunner Reese (Gideon Emery) again.

Upon reaching the scene, the survivors encounter additional members of the Whiskey Company: Chief Cole, Deputy Chief Lewis, Chief Engineer Carter (Jason Harris), Heavy Weapons Officer Jericho (Christopher Judge), Weapons Specialist Logan (William Fichtner), Doctor Parker (Joe Shua Gomez), Technician and Pilot Shepard (Donnie Wahlberg), and Soldiers Foster (Jon Curry) and Gonzalez (Lombardo Boyar). Turok is tasked with finding the ship's communications unit, encountering Kane's deputy chief John Grimes (Sean Donnellan), and later another survivor, Cowboy (Timothy Olyphant), who was shot and wounded by Grimes. The Whiskey Company comes to Cowboy's aid. And Turok returns to the search with Foster and Gonzalez. After Foster is killed by MG gunmen, Turok and Gonzalez retake the communications unit, but Gonzalez is grabbed by a large Tyrannosaurus Rex. Turok attacks the Tyrannosaurus but is knocked unconscious.

Turok awakens and travels back to the communications unit, where Slade accuses him of betraying his teammates again. Cole (Mark Rolston) is killed in a surprise attack by Grimes, and MG forces attack the area. Whiskey Company fends off the attack, but Parker and Lewis are killed, and the communications unit is destroyed. Logan assumes command. He orders Turok, Slade, and Carter to investigate the distant lights.

Upon reaching the abandoned base, the trio discover that Kane has been poisoned by a powerful neurotoxin from a scorpion-like creature, as well as the location of a ship in a substation near the MG base. The same creature attacks them, killing Carter, while Turok and Slade fall into a cave system. Turok saves Slade from a large tentacle creature, earning Slade's respect. They escape the cave and join forces with the remaining members of the Whiskey Company.

An approaching MG patrol causes Logan to accuse Turok of betraying the Whiskey Company, but Slade protects Turok. The patrol accidentally finds them and Logan is killed. The group fights their way to the substation, and Jericho sacrifices himself to allow them access. They find the ship abandoned and travel to the MG base to search for another. At the entrance, Reese is killed by Grimes from a distance. They make it inside, aided indirectly by the Tyrannosaurus that Turok had previously fought. The group also learns that Kane has developed a deadly gas from the Scorpion creature. To keep the bioweapon from leaving the planet, Turok plants a bomb on the base's generator. Kane captures them and kills Cowboy, but Turok detonates the bomb, killing Grimes.

Turok, Shepard, and Slade escape the crumbling base. They reach another ship, but Turok refuses to leave until Kane is dead. Turok shoots down Kane's ship, and they engage in a knife fight, resulting in Turok killing Kane. A Tyrannosaurus confronts Turok, who thrusts a grenade into its eye, killing it. Turok is carried aboard Shepard and Slade, and they set off on a path back to Earth.
