This is after saving Ilia, after the 2nd King Bulblin fight. While Link is in Kakariko following Queen Zora, it seems as though it creates a flag for BiTE as well. While following Queen Zora, it seems apparent that it creates a trigger/key for the 2nd portion of the first King Bulblin fight when BiTE is used with this portion of the game.

Kakariko seems to hold something special for BiTE, but whether or not it will do anything else will remain questionable until further testing.
