Edit: This method seems to work on the PC version only. I tested it on the Xbox version and it seems impossible though it may be just a timing issue with the exiting and entering of vehicles and releasing the side mission trigger button (the look behind button does nothing on the Xbox version).
This is by far the easiest method to complete this side mission, especially if the gangs are hostile towards you. You must use the "Taxi Driver" and car crusher glitches to make your vehicle invulnerable to physical damage and bullets (it won't remain bulletproof if you store it in a garage). It will turn the Ambulance into a ghost car. As a result you won't be able to run patients over and you can drive through buildings to get to your destination faster.
