This final side mission is unlocked after Last Stand, the final main storyline mission. If you're going for 100% completion, you can skip this side mission entirely or technically any other side mission aswell, but only if you obtain a drugs empire site business before the storyline mission Hostile Takeover. Doing so, will give extra percentage added, making it possible to 100% before Skywolf.

You could also choose what side mission you want to skip when you got that extra percentage, for example a hard side mission you don't want to complete, just skip it with this trick. You still get 100%.
