Here is the method I used for grinding puzzle pieces in Kirby Star Allies, which as far as I'm aware is the fastest method. This method nets you 21 puzzle pieces in about 6 minutes. You also get to suplex a lot and that's always nice.

Castle Dedede has a hidden room which contains tons of puzzle pieces, but you have to go through a series of timed challenges and a mini boss to get to them. By taking Marx as a Dream Friend and riding him, you can use his air dodge to go against the wind current and bypass all that. Apart from that, Castle Dedede also has a ton of puzzle pieces lying around that are quick to get.

I also bring Magolor as his Ultra Sword can hit all the switches in the room where you're split up (make sure he's in slot 3), and he's generally useful on the Dedede fight. Susie is brought along too as her copter can sometimes speed things up.
