I saw someone playing this mod with this challenge setting and I thought it'd be fun to try.

SpiderMod 0.9 is a comprehensive mod for Binding of Isaac that allows for mass customization of the game. This boosts replay value even more because you can create your own challenges and runthroughs

Universal Item Pool changes the rules of where items drop. Typically Boss Rooms, Deal Rooms, Shops, Item Rooms, Libraries, etc have their own specific item pools. This mod changes those rules to allow items to show up anywhere in the game. This would make for some interesting runs.
