With the earth crystal out of the way, we can focus on the vampire sidequest.
To unlock the door you will need to beat the dragons at the obelisks all over the map. There is 1 located in every region.
After collecting them all you are permitted entrance into the castle.

Inside the castle there is quite a bit of dialogue in which DeRosso explains about the Orthodoxy and his own past. This part contains some spoilers so you may wish to avoid it.

At the top of the tower Lester challenges you to a fight.
He is capable of dealing incredible amounts of damage if you let him. Graviga can do a decent amount of damage but it tends to miss quite often. Energy burst will do a lot of damage but it can be negated with the Templar's Rampart. The attack which you have to watch out for is Bone crush. Bone crush's damage differs depending on how much damage you have. If you are fully healed then it will deal 0 damage, but it can increase VERY quickly. Always have your health full just to be safe. Rampart is an incredibly helpful ability for this fight since it negates both Energy burst and Bone Crush.
