This video shows you the steps where even with a new character, you can get over 45,000 Runes an hour without having to do anything but wait. You could even put down the controller for about 5 minutes at a time and go do something else to come back and have extra Runes.

You could also aggro additional dino-dogs to join the fight but this trick is a lot riskier.
