TaqMan PCR for Accurate Diagnosis of Tick-Transmitted Pathogens in Cattle | Research Study from Kerala, India

🔬 About This Study:
This video discusses a research study that utilized TaqMan-based qualitative Real-Time PCR (qPCR) for the accurate and confirmatory diagnosis of multiple tick-transmitted pathogens affecting the bovine population in Kerala, India. Conventional microscopy-based methods often fail to identify pathogens at the species level, especially when they have similar infective stages. This study highlights the advantages of molecular diagnostics in detecting co-infections and secondary pathogens that impact cattle health.

Sample Collection: 60 peripheral blood samples (EDTA) were randomly collected from five private cattle farms in Wayanad & Thrissur districts, Kerala.

Pathogens Detected: The study identified Theileria spp., Mycoplasma haemocanis, and Pasteurella multocida using highly sensitive TaqMan qPCR.

Ct (Cycle Threshold) Values:
Theileria genus: Avg Ct = 12.79 (Range: 4.1–36.19)
M. haemocanis: Avg Ct = 22.42 (Range: 2.66–37.6)
P. multocida: Avg Ct = 20.20 (Range: 2.74–35.72)
(Lower Ct = Higher pathogen load in initial sample)
P. multocida as a Secondary Pathogen: Found to cause severe issues due to immune suppression by primary tick-borne infections.

Vaccination Strategies: Co-infections may interfere with seroconversion & duration of immunity, requiring tailored vaccination programs.
Need for Advanced Diagnostics: Microscopy alone is insufficient; molecular methods like TaqMan PCR improve detection accuracy.
