I usually don't run Desperado or the Rifle but I wanted to change it up. What a fantastic combo having Lysate Cell so you can keep firing the Desperado. Eventually you can just one shot everything and keep firing to continue building stacks. This is the type of build that snowballs on the stage and then you have to rebuild it all over the next stage. This is one of my slower runs as well because I was just spending too much time on each stage shooting lesser mobs, still was fun though.

Aiming for the flying mobs, beetles, are great for building up the stacks. The only problem with the last stage is the little orb don't die in 1 shot and the on-proc hit stuff will usually kill them (ie: bands). So trying to stack on the last stage was a little difficult until around 13 or so and then I could continue one shotting them. I probably shouldn't have picked up Ceremonial Daggers, that kinda defeated the purpose of killing everything with Desperado.
