In Demon Slayer: Hinokami Chronicles chapter 3, Tanjiro Kamado and his sister Nezuko find themselves in Asakusa, Tokyo, embarking on their next mission. As they investigate the town, Tanjiro suddenly detects the unmistakable scent of the powerful demon king, Muzan Kibutsuji. Determined to confront him, Tanjiro becomes distracted when Muzan turns a human into a demon right before his eyes. This diversion allows the demon king to escape, leaving Tanjiro frustrated.

However, during his search, Tanjiro encounters a mysterious and kind-hearted woman named Tamayo, who reveals herself to be a Demon Doctor. Intrigued by Nezuko's unique condition as a demon who hasn't consumed human blood and yet remains peaceful, Tamayo seeks the opportunity to study her. She believes Nezuko's blood could hold the key to developing a cure for demons, an ambitious goal she has been pursuing. Together, they work to sedate the newly-turned demon without attracting any unwanted attention, all the while forming a potentially crucial alliance in the quest to find a remedy for demonkind.

Lastly, Tanjiro and Nezuko face off against Sasamaru and Yahaba in a final showdown. Tanjiro fights Sasumaru by himself for the first phase of her fight. During the second phase nezuko takes Sasumaru on and Tanjiro fights Yahaba, who's been enhancing Sasumaru this whole time. It ends in the two sliced heads and Doctor Tamayo getting the demon blood she needs to further her Demon Cure research. Tanjiro and Nezuko head off, to the mountains.
