Once you reach 250,000 Balls, go to the area shown in the Map.
Like all Femme Fatales, Isabelle can be found in this location before the mission is unlocked. However, when talking to her the conversation will play out as usual until the last line in which she will insult or reject Tony in some way (which may differ while the rest of the conversation lines go on again). Generally continuing conversation will go through a few random lines until the conversation begins again, same as before including with a different rejection line.
Once you have the requisite number of Balls, the conversation will again be repeated as though it never prior occurred, but ending with the same acceptance line.
Oddly after this she will be totally non-responsive to Tony, and will not even react to gunfire. She can be hit with fists or with a car but will simply get back up and roam around after as if nothing happened. If you leave the location and return she will be gone and will never return here. This non-responsive behaviour will also continue in the Mansion if you use the exploit I've found here (place an Exotic where she is supposed to stand, which makes her walk around trying to find her way to that blocked location). Use this exploit before recruiting the Femme Fatales if you'd rather them walk around silently instead of stand around with nothing meaningful to say as they do normally.
This encounter gives you a Stamina upgrade regardless of which order you approach the Femme Fatales in.
