When Desmond Miles enters the sanctuary of the Auditore Villa with Lucy Stillman, he sees an apparition of an old Ezio Auditore and Desmond wonders why Ezio returned to the villa after the attack on Monteriggioni. Arriving at the secret entrance to the Sanctuary behind the bookshelf in Mario Auditore's study, Desmond then uses his eagle vision to find a mark on the wall with numbers: 1419, 1420, 1421. As Desmond's group will later find out: It is a 72 marking! The number 72, which refers to the 72 names of God, played an important role for the Assassins.

Conclusion: Ezio Auditore returned to the villa in 1506 after sealing the Apple of Eden in the Colosseum Vault to leave the 72 mark as a clue for the password to open the Vault in Rome to Desmond or any assassin who has the eagle vision ability. 🧐 🙂
