Genocide Run involves killing every possible NPC in Dark Souls within a single playthrough. This does not include characters such as Frampt, who cannot be killed. It also does not necessarily mean killing NPCs as soon as they are first encountered, as it is sometimes necessary to follow a character's questline for another character to appear.

NPCs in Part 6:

-Eingyi and Quelaag's sister: they can both be killed at any time you like, but Quelaag's unnamed sister is a firekeeper so the bonfire will be extinguished if you kill her. you will also need her to upgrade your estus flask, so for this reason it is best to wait until the end of the game before you kill either of these NPCs.

-Oswald of Carim: he can be killed at any time but he also sells purging stones so it's probably best to wait until the end of the game.

-Crestfallen Warrior: he can be killed at any time, although i suggest waiting until you've got a decently upgraded weapon.
