Genocide Run involves killing every possible NPC in Dark Souls within a single playthrough. This does not include characters such as Frampt, who cannot be killed. It also does not necessarily mean killing NPCs as soon as they are first encountered, as it is sometimes necessary to follow a character's questline for another character to appear.

NPCs in Part 10:

-Gwynevere: being an illusion, she could be considered optional for a genocide run. however, there are two other NPCs who will only appear if the illusion is dispelled. for this reason it is actually necessary to "kill" Gwynevere, although this will also antagonise the Darkmoon Knightess so i suggest you don't do this until your Estus Flas is fully upgraded to +6

-Siegmeyer of Catarina: you must exhaust his dialogue when he returns Firelink Shrine before his questline can progress any further. he will then appear in Blighttown when you head down there, and you must agree to give him Purple Moss Clumps. you will need to have at least 3 in your inventory to be able to do this.

-Kirk, Knight of Thorns: this invader has three scripted invasions throughout the game and you must kill him all three times in order to properly hollow him. the second is in the Demon Ruins, although this invasion will not occur before you've placed the Lordvessel, neither will it occur after you've killed the Firesage Demon.

-Patches: ordinarily he will attempt to scam you at this point, but i had already visited the Tomb of the Giants in part 5 of this series. Patches can be killed immediately if you like, but will eventually become a merchant if you spare him for now.

-Vince, Nico, and Rhea of Thorolund: ordinarily these characters would be found in the pit that Patches kicks you into, but i had already killed them in part 5 of this series.

-Paladin Leeroy: this invasion will only occur before you've beaten Gravelord Nito.
