To use Push to Talk on a controller in Fortnite, follow these steps:

Launch Fortnite and navigate to the game Settings.
Click on the Speaker icon at the upper part of your screen.
Shift the toggle next to Voice Chat to the On position.
Optionally, adjust the sound quality, subtitles, and other settings.
Make sure to assign the push to talk to a button on the controller layout otherwise it won't work.
