I personally found this easiest to complete by equipping the Throwing Knife and heading to Hanging Dog Ranch, far West of Valentine. Make sure to purchase Throwing Knives from a Fence or the Trapper, and then ride Northwest along the river until you reach the cow pen.

Use Dead Eye to aim for the head of the first farmhand on your left near the fence, then kill the other two slightly past him straight ahead. If you miss one, as you turn back to leave the way you came, there will be another enemy that jets out from behind the outhouse that you can kill to complete this challenge as well!
