Location: Fountainhead Palace
This can only be accomplished once you have unlocked the "Puppeteer" Ninjutsu by defeating the Folding Screen Monkeys boss at the end of the Senpou Temple Mt. Kongo region. Additionally, you must have progressed through the Sunken Valley region in order to encounter the Great Serpent. To begin, make your way to the Senpou Mt. Kongo idol, which is the first idol in that region. From there, descend the cliff on the right side where you will find a lone enemy near a lever attached to a kite. Execute a backstab on this enemy and use the Puppeteer skill on him. He will then operate the lever to manipulate the kite and keep it in place. Proceed through the monk forest, pass the prayer room, and continue to the next area where you will spot a large tree on the right side. From that tree, you can grapple onto the kite and soar to the adjacent area. Descend the platforms while utilizing tree branches as anchors, until you reach the Sunken Valley Cavern idol. From there, follow the path to a dead-end where you can carefully balance on a wooden beam, and below you, the serpent will be present, but only if you have already made progress in the Sunken Valley region.
Strategy: This "true" version of the Corrupted Monk possesses three lives. Fortunately, we can swiftly eliminate his first two lives through stealth kills from the trees above. Upon entering the boss area, grapple onto the tree branch on the right, then onto the second one on the right, and finally onto the third branch positioned behind the boss. If you act swiftly, he won't notice your presence, allowing you to descend upon him for a stealth kill. Immediately return to the trees, and when he transitions into his second phase, execute another stealth kill from the middle tree branch (despite the boss turning invisible, he stands in the middle of the bridge, where you can drop down on him—be sure to act quickly). Now, he is left with just one life. Utilize firecrackers to distract him, launch regular sword strikes from behind, follow up with another firecracker, and repeat the process. Additionally, consume Ako's Sugar for increased attack power and Ungo's Sugar for reduced damage taken. Using approximately eight sets of firecrackers should suffice to deplete his final life. Alternatively, you can evade his spinning attacks by swiftly moving away from him, then close in to land a single hit, and repeat the pattern. The Mortal Blade Combat Art is also effective when baiting him like this, as it deals unblockable damage.
