This is a remake of an older video (https://www.youtube.com/watch?v=nNukgmW_O5k).

Instant enters:
First two junctions: right, down-right
Floor 1 Door: right
Floor 2 Door: left
Floor 3 Door: right (shout outs to Nate for setting me straight on this one)

Camera angles are slightly different for each character, meaning these instant enters may not work for any other character besides Con.

There are three clips here, though the only real difference is the first floor.

Route 1 (0:00)
Mach Speed is used on the first floor, and the first Smile is dodge rolled. It would naturally be faster to dodge roll it without scanning, but I never trust myself with it. Floors 2 and 3 are easy, running past enemies without issue. If I recall correctly, Simna was the one who had the idea of turning around on floor 2. I did test Mach vs. No Mach on floor 2, but they ultimately broke even, and it really isn't worth it.

Route 2 (0:28)
Mach Speed is still used on the first floor, but the first Smile is tanked. It is very close in time to the first route (maybe slower by just a few frames), and is arguably easier, though I find the junction right after the tank to be a bit finicky.

Route 3 (0:57)
For the sake of completion, I included a clip without Mach on the first floor, which is slower, but really not by much. It can be useful if you're not feeling confident with the Mach Dodge.

On Killer8, Route 1 would still be the fastest, but it would be pretty risky, obviously. I would go for Route 3 for safety. On Normal, the blood route is perfectly fine by this point, but on Killer8, killing the Backside on the second floor is recommended for a little extra blood for the Ayame fight.
